Is it possible to run index.php in the root directory or do we have to settle for setting up the domain with a 301 error?

PostNuke allows install of index.php in the root but I have done so much work with my phpnuke I don't want to start all over again if possible.
Any replies would be grapely appreciated. I've been searching for 3 days now with no replies anywhere.
